The Radioactive Spider was a spider mutated by being accidentally exposed to fatal doses of radiation during a public demonstration of the safe handling of nuclear waste. It lived only for a short time after its mutation, and was responsible for biting Peter Parker and Cindy Moon, infecting them with its venom which in turn altered their DNA, allowing them to develop arachnid abilities. The spider died shortly afterwards.

- Some sources claim that the radioactive spider was actually an avatar controlled by the Great Weaver.
- A number of adaptations have replaced the radioactive spider with a genetically-engineered one. These include the cartoon series The Spectacular Spider-Man and Ultimate Spider-Man, and the live action films Spider-Man and The Amazing Spider-Man.
- In its first appearance, the comic's narration refers to the radioactive spider as an insect, even though spiders are actually arachnids.
